The PathHunter eXpress XBP1 Nuclear Translocation Assay provides an easy-to-use cell based assay to study drug candidates affecting the IRE1/XBP1 pathway. This kit assesses ligand (e.g. thapsigargin) based pathway activation via detection of resulting XBP1 nuclear translocation. This product uses EFC technology. The included cell line overexpresses ProLabel-tagged XBP1 and nuclear localized EA. Rat Active tPA ELISA Kit - This rat tPA activity assay is intended for the quantitative determination of active tissue plasminogen activator (tPA) in rat plasma and other biological fluids. For research use only. Strip well format. Reagents for up to 96 tests. Functionally active rat tPA will form a covalent complex with the biotinylated human PAI-1 which is bound to the avidin on the plate. Only free active tPA will react with the PAI-1 bound to the plate. After appropriate washing steps, polyclonal anti-tPA primary antibody binds to the captured tPA. Excess antibody is washed away and bound primary antibody is then reacted with the secondary antibody conjugated to horseradish peroxidase. Following an additional washing step, TMB substrate is used for color development at 450nm. The amount of color development is directly proportional to the concentration of active tPA in the sample.

Free and complexed tPA will bind to the plate. After appropriate washing steps, monoclonal anti-rat tPA primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with the peroxidase conjugated secondary antibody. Following an additional washing step, TMB is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of rat tPA. Color development is proportional to the concentration of tPA in the samples.
